Rabbit Polyclonal antibody to Grp78 (78 kDa glucose-regulated protein)When Chinese hamster K12 cells are starved of glucose, the synthesis of several proteins, called glucose-regulated proteins (GRPs), is markedly increased. Hendershot et al. (1994) [PubMed 8020977] pointed out that one of these, GRP78 (HSPA5), also referred to as ＇immunoglobulin heavy chain-binding protein＇ (BiP), is a member of the heat-shock protein-70 (HSP70) family ａnd is involved in the folding ａnd assembly of proteins in the endoplasmic reticulum (ER). Because so many ER proteins interact transiently with GRP78, it may play a key role in monitoring protein transport through the cell.[supplied by OMIM]

• Non-transfected (–) ａnd transfected (+) HepG2 whole cell extracts (30 µg) were separated by 7.5% SDS-PAGE, ａnd the membrane was blotted with Grp78 antibody diluted at 1:10000. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody.

Application

• Grp78 antibody detects Grp78 protein at cylasm by immunofluorescent analysis. Sample: A431 cells were fixed in 4% paraformaldehyde at RT for 15 min. Green: Grp78 protein stained by Grp78 antibody diluted at 1:500. Blue: Hoechst 33342 staining.

• Various whole cell extracts (30 µg) were separated by 7.5% SDS-PAGE, ａnd the membrane was blotted with Grp78 antibody diluted at 1:10000. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody.
