吴桂英出生于1966年，长期在北京市工作，曾任北京市计划委员会副主任、北京市发展和改革委员会副主任，朝阳区委常委、副区长。
2012年7月，任朝阳区委副书记、代区长，同年11月当选朝阳区区长；2015年9月，任朝阳区委书记。
2018年1月，调任湖南省副省长；2021年2月，任湖南省委常委、长沙市委书记至今。
